A corneal-PAMPA-based in silico model for predicting corneal permeability

چکیده
• QSPR models were developed based on in vitro corneal permeability measurements of 189 compounds. Partial least squares regression was applied for the model building. Classical molecular descriptors and ECFP fingerprints used. Corneal membrane retention values predicted by models. The capability to predict physicochemical parameters has always been a desirable objective ophthalmic drug development. However, previous work limited cases where either diversity compounds used lacking or performance poor. Our study provides extensive quantitative structure-property relationship (QSPR) predictions. involved diverse Preliminary analysis data showed that there is no significant correlation between corneal-PAMPA (Parallel Artificial Membrane Permeability Assay) other pharmacokinetically relevant silico transport like Caco-2, jejunal blood-brain partition coefficient (logBB). Two different developed: one retention, experimental data. producing models, which contained classical combination. A complex validation protocol (including internal external validation) carried out provide robust appropriate predictions values. Both had an overall fit R 2 > 0.90, including -values not lower than 0.85 runs, quick accurate set

Development of Novel In Silico Model to Predict Corneal Permeability for Congeneric Drugs: A QSPR Approach
This study was undertaken to determine in vivo permeability coefficients for fluoroquinolones and to assess its correlation with the permeability derived using reported models in the literature. Further, the aim was to develop novel QSPR model to predict corneal permeability for fluoroquinolones and test its suitability on other training sets. The in vivo permeability coefficient was determined...
متن کاملPrimary Corneal Keloid
Purpose: To report the clinical presentation, histopathologic findings, and surgical treatment in a series of cases with primary corneal keloid. Case Reports: Six Iranian patients (a father and two sons, a mother and her daughter, and one isolated male patient) without any history of corneal trauma or systemic diseases were clinically and histologically evaluated for a slowly-growing, white opa...
متن کاملCharacterization of human corneal epithelial cell model as a surrogate for corneal permeability assessment: metabolism and transport.
The recently introduced Clonetics human corneal epithelium (cHCE) cell line is considered a promising in vitro permeability model, replacing excised animal cornea to predict corneal permeability of topically administered compounds. The purpose of this study was to further characterize cHCE as a corneal permeability model from both drug metabolism and transport aspects. First, good correlation w...
متن کاملPrediction of Corneal Condition After Corneal Ring Implantation in Keratoconus Patients
Background: Keratoconus is a common complication among corneal defects. As a result of expeditious and extensive progress of medical science in recent decades, corneal ring implantation has turned into a successful surgical procedure to correct the vision of Keratoconus patients; however, selecting the right patient is essential in the success of the operation. The prediction of corneal conditi...
متن کاملParacellular permeability of corneal and conjunctival epithelia.
The paracellular permeability of normal rabbit cornea and conjunctiva was studied in vivo and in vitro. After intravenous administration, horseradish peroxidase was found to percolate to the intercellular space of conjunctival epithelia and was restricted by the tight junctions of the superficial epithelium. Only minimal tracer was present in the limbus and cornea. The difference between cornea...
